Impact of vitamin D Supplementation on Glucose Metabolism in Type 2 Diabetes

Abstract:
Type 2 diabetes mellitus (T2DM) is a chronic metabolic disorder characterized by insulin resistance, hyperglycemia, and progressive pancreatic beta-cell dysfunction. In this context, vitamin D deficiency has emerged as a potential risk factor that may contribute to poor glycemic control. This review aimed to analyze current scientific evidence regarding the effects of vitamin D supplementation in individuals with T2DM and its relationship with clinical parameters such as fasting glucose, glycated hemoglobin (HbA1c), and the HOMA-IR index. A systematic search was conducted in international scientific databases, including studies published between 2010 and 2025. Findings suggest that vitamin D supplementation, particularly in patients with deficiency, can significantly improve insulin sensitivity and certain glycemic indicators. The most favorable outcomes were observed with high-dose vitamin D interventions (≥ 4,000 IU/day or ≥ 50,000 IU/week) administered for short durations (up to 12 weeks). However, the effects vary depending on dosage, treatment duration, and patients’ baseline nutritional status. Evidence indicates that vitamin D may play an adjuvant role in the nutritional and metabolic management of T2DM, highlighting the importance of evaluating its status within comprehensive clinical care.
