Standardization Of In-Vitro Root Induction Protocol for Pomegranate (Punica Granatum L.) Variety Solapur Lal

Abstract:
Pomegranate (Punica granatum L.) is a commercially important fruit crop, but its propagation is hindered by poor rooting. This study investigated the effects of indole-3-butyric acid (IBA) and α-naphthaleneacetic acid (NAA) on in vitro root induction in the biofortified pomegranate variety Solapur Lal. Proliferated nodal explants were sub-cultured on a 3 basal medium (MS, WPM and DKWJ) supplemented with Activated charcoal (500mg/L), PVP (500mg/L), 0.5 mg/L IBA and 0.5 mg/L NAA, alone and in combination. Among the treatments studied, W2 (without IBA and NAA) exhibited the least days required for root initiation (14.33 days), maximum root number (1.333), and root length (5.333 cm). Additionally, W2 showed a good greenness index (5) and net shoot growth (2 ± 0.2 cm). These results suggest that the medium with WPM with activated charcoal and PVP is optimal for in vitro root induction in pomegranate cv. Solapur Lal, and can be used for large-scale propagation of this commercially important cultivar.
