Chapter Chapter 4 in Quantitative Methods in Pharmaceutical Research and Development, 2020, pp 143-179 from Springer

Abstract: Abstract In this chapter readers will be introduced to some of the biostatistical methods used in the analysis and interpretation of Pharmacoepidemiological data. We present the measures used to estimate the frequency of a disease and how these are used to assess an exposure-effect association in Pharmacoepidemiology. Then, we describe how to take into account common issues in observational epidemiology, such as bias, confounding, and interactions (e.g., exposure by covariate interactions), in order to establish a clear causal link between the exposure and the effect. Finally, we present the common statistical methods used to address confounding and interactions (effect modifications) analytically.
