Abstract:
Abstract The complex kinetics of reversibly photoswitchable fluorescent proteins (RSFPs) is exploited in biological imaging. We analyze the fluorescence evolution of RSFPs submitted to two-color illumination at different light intensities appropriately chosen to change the rate-limiting steps of photoswitching. Starting from a reasonable seven-state photoswitching mechanism, we use the steady-state approximation to obtain simple reduced mechanisms associated with a small number of characteristic times. The comparison with experimental results for the RSFP Dronpa-2 provides previously unknown kinetic information on photoswitching.
