Kinetic and Mechanistic Aspects of Pregabalin Oxidation with Alkaline Potassium Permanganate

Abstract: The kinetics of the oxidation of pregabalin by potassium permanganate in basic medium, maintaining a constant ionic strength of 0.1 mole dm−3, were investigated using spectrophotometry ally at 25°C. It was determined that the stoichiometric ratio between permanganate ion and pregabalin in alkaline conditions is 1:2. The kinetics of the reaction between Pregabalin and the permanganate ion has been observed to follow pseudo first-order behavior. A less than unit order dependence of the reaction on both Pregabalin and [OH−] was noted. There was no significant impact on the reaction rate due to increased ionic strength. The primary products were characterized using M.P. and IR spectral analysis. The rate of reaction was unaffected by the ionic strength and dielectric constant. The addition of certain products did not meaningfully influence the reaction rate. The rate law for the oxidation reaction has been determined. The reaction constants and mechanism have been calculated. The activation parameters linked to the rate-determining step of the reaction, as well as the thermodynamic values of the equilibrium constants, have been evaluated and discussed.
