The effect of crude glycerol impurities on 1,3-propanediol biosynthesis by Klebsiella pneumoniae DSMZ 2026

Abstract:
This study aimed to evaluate the ability of Klebsiella pneumoniae DSMZ 2026 to biosynthesize 1,3-propanediol (1,3-PD) during batch fermentations when pure glycerol was supplemented with 10 g of unpurified biodiesel-derived glycerol. The crude glycerol (CG) fraction was obtained at laboratory scale from waste cooking oil. Cell viability, biomass, metabolites production, and substrate consumption were investigated. After 12 h of batch cultivation, 18.81 g/L 1,3-PD (yield 0.42 g/g, productivity 1.57 g/L/h) was reached when only pure glycerol was used, having a starting concentration of 43.5 g/L. When 10 g of unfiltered CG was added to the fermentation media that contained pure glycerol, 9.69 g/L 1,3-PD (yield 0.21 g/g, productivity 0.80 g/L/h) was obtained after 12 h. This work underlines the inhibitory potential of unfiltered GC fraction over the 1,3-PD production by K. pneumoniae DSMZ 2026.
