Abstract:
Ion pumps: the outside story P-type ATPase ion pumps are found in virtually all cell membranes pushing ions across the membrane against their concentration gradients. Structural studies have made tremendous advances in recent years and a lot is now know about what the insides of ion pumps look like, but it is still not clear how they collect ions on one side and release them on the other. Now by using the marine toxin palytoxin to uncouple the gates that control access to the pump's ion translocation pathway of Na+,K+-ATPase, thereby transforming the pump into a cation channel, Nicolás Reyes and David Gadsby have obtained a novel view of the ion pump in action. At the extracellular side, the ion pathway begins as a deep and wide vestibule, which abruptly narrows and leads to a cation selectivity filter made up of acidic sidechains. Strikingly, reversing the negative charge at just one of them converts the pathway from cation- to anion-selective.
