Gadd45a promotes epigenetic gene activation by repair-mediated DNA demethylation

Abstract: DNA methylation reversed It is increasingly appreciated that DNA methylation, a ubiquitous regulatory mechanism, can be reversed by a process termed 'active demethylation', but its underlying mechanism is poorly understood. Advance in the DNA demethylation field is hampered by a lack of knowledge about the key regulators that could serve as a starting point for unravelling this process. Barreto et al. now report the discovery of one such regulator, Gadd45a, or growth arrest and DNA damage inducible protein 45-alpha. Gadd45a, shown previously to be a target of the p53 and BRCA1 tumour suppressor genes, is acting here by promoting DNA repair to erase methylation marks and thus relieve gene silencing.
