Two enzymes bound to one transfer RNA assume alternative conformations for consecutive reactions

Abstract: Enzymes sharing space on tRNA Synthesis of Glu-tRNA(Glu) and Gln-tRNA(Gln) begins with a common reaction, in which the glutamyl-tRNA synthetase (GluRS) glutamylates both tRNA(Glu) and tRNA(Gln). The Glu-tRNA(Gln) is then specifically converted to Gln-tRNA(Gln) by an amidotransferase. Takuhiro Ito and Shigeyuki Yokoyama have now solved the structure of a bacterial macromolecular 'glutamine transamidosome' complex containing tRNA(Gln), GluRS and the trimeric amidotransferase GatCAB. The structure explains the hand-off mechanism between the two enzymes, in which only one can assume a productive state at any time, and suggests that there is an intermediary state in which neither enzyme is productive.
