Abstract:
Understanding a multi-subunit complex The large size of the anaphase-promoting complex or cyclosome (APC/C) and its low natural abundance combine to make quantitative structural and mechanistic studies of the complex difficult. Two linked papers take on the challenge. Schreiber et al. have reconstituted the entire APC/C complex and, in combination with structural studies, provide a pseudo-atomic model for 70% of the complex. These results contribute towards a molecular understanding of the roles of individual subunits in APC/C assembly and their interactions with co-activators, substrates and regulatory proteins. Da Fonseca et al. present a cryo-electron microscopy structure of APC/CCdh1 bound to a D-box peptide substrate. The structure provides insight into the recognition and catalytic mechanism of APC/C substrates.
