Abstract:
Catalytic activity of ribosomes The catalytic centre of the ribosome is composed solely of RNA. This means that it lacks the varied side groups found in proteins that can assist in catalysis, so it was thought that its functions were merely to bring messenger RNA and charged transfer RNAs into close proximity. Not so. Analysis of the kinetic isotope effect at five positions within the reaction centre of a peptidyl-transfer RNA mimic shows that the ribosome also enhances catalysis of peptide-bond formation by modifying the rate-limiting transition state. This finding has implications for work on the origin of life and the transition from an RNA world.
